Here is how to access Ranni’s Great Rune through the Carian Study Hall in Elden Ring. On the first visit, the Divine Tower of Liurnia is inaccessible, and you must progress Ranni’s questline to reach the tower. In the process, you will get your hands on Ranni’s Great Rune.
Your first visit to the Carian Study Hall

When you reach the interior of the Study Hall, you’ll have to battle with Preceptor Miriam. Killing her can be tricky because she keeps attacking you from afar, and she teleports back and forth. Kill her and explore the area. But in doing so, you’ll understand that you have to stand on a switch on an elevator. But there is no way to reach it because it’s upside down. You’ll have to postpone trying to explore this area at that point.

Ranni’s Questline
Ranni instructs you to obtain the Secret Treasure of Nokron and present it to her, as a part of her questline.
After defeating General Radahn, a crater will appear in the Mistwood region of Limgrave. It will be marked on your map with a red marker. Entering this crater will lead you to the underground city of Nokron. Do keep in mind that you will have to complete the necessary steps in Ranni’s questline at this point in order to trigger certain events.
Anyway, by exploring this uniquely beautiful city, you will reach a location with a chest that contains the Fingerslayer Blade. This is the secret treasure of Nokron. Take this to Ranni at Ranni’s Rise and she will thank you. And she also rewards you with the Carian Inverted Statue. This is what we need to freely explore the Study Hall.
Going back to the Carian Study Hall
Go back to the Study Hall and interact with the Globe on the desk.
Now you have revealed the secret areas of the Study Hall. When you enter it, you’ll notice how it’s inverted. It’s uniquely beautiful, in a bizarre way. Here, you’ll once again have to defeat Preceptor Miriam. There will also be backup, so do your best. Keep an attentive eye and you’ll notice some items around. Now you’ll be able to climb on top of some wooden beams (which you would’ve been on in your first visit). But now that the Study Hall is inverted, you will be able to jump on the lift.
Now it’ll be a trivial matter to reach the Divine Tower of Liurnia. However, you will have to face a Godskin Noble enemy. Kill him and get on the lift to reach the Tower. At the top, you’ll find the Cursemark of Death on a burned body. This burnt body is in fact, Ranni’s. It was her body before she became a doll.
The Cursemark of Death is required for an ending in the game, and to spawn a boss.
